Abstract Bachelor Project FBT 2018-2019: Evaluation on the efficacy of active chlorine for disinfection of drinking water for animals in the context of the ‘Biocidal Products Regulation’
 
To release a biocide, registration is necessary according to the Biocidal Products Regulation (BPR). To comply regulations, a field trial – or phase 3 test – has to be performed. (European Chemicals Agency, 2018) In this study, a product containing active chlorine for disinfection of drinking water for animals is evaluated.
 
To analyze if the product is working properly, water samples were taken at the end of the lines in a poultry house. Herefore, two stables were compared: a treated stable (S1) and an untreated stable (S2). There was only a small difference between these two stables. S1 exceeded the norm one time for bacteria. S2 crossed the norms for drinking water fifteen times on all bacteria, seven times on yeasts and moulds and six times for Coliforms.
 
This field trial had already been done at another farm and showed a higher difference between the stables. Even though the results of stable 1 are better than stable 2, a higher difference between the two stables was expected. It is advisable to redo this field test and to investigate why the difference is not as outspoken as in previous field trial
 
Samenvatting eindwerk 2012-2013: Development of a protocol on residues analysis of disinfectants after application on surfaces
Use of disinfectants requires a good view on the quantity of residue that stays on the surface after application. Disinfectants are used in the food industry on equipment which comes directly into contact with food. They are also used in the agriculture on surfaces where animals stay. It’s important that it can be ensured that not too much residue of the product will be rejected in the environment and be toxic for human and animal. It’s a very important aspect for the government because it must be ensured that the product has no toxic values. Therefore, residue analysis should certainly be carried out.
The target of this project is to set up a protocol that can be used to test how much residue disinfectants leave after application on a surface. This protocol is developed and meant for residue determination of products that are applied on stainless steel.
The developed method must be validated for each active substance for which the protocol is used. The three active substances that were investigated are hydrogen peroxide, ethanol and glutaraldehyde. These substances are frequently used in several disinfectants. The validation of the developed method is performed to ensure the reliability of the obtained results. Ensuring the reliability was one of the most important subjects. The main problem was the repeatability of the results. Mainly ethanol and glutaraldehyde had problems with this parameter.
After the validation, a ‘profile’ is created for the three active substances. This profile is set up to know how the substance behaves in different conditions occurring in the field. The accumulation and saturation of the substance are investigated. Also the quantity of rinsing water required to remove all residue is examined. The results can be used as additional information for residue tests of disinfectants in which the active substance is used.
Three disinfectants, CID CLEAN, ALCO CID A and Biocomplet plus, containing respectively hydrogen peroxide, ethanol and glutaraldehyde, are investigated on their quantity of residue after application on inox. The conditions applied are those used in the field; like the application quantity, rinsing quantity and contact time. A report of the results is made and will be sent to the government. The obtained residue values for the three disinfectants are 11,7 mg/m² hydrogen peroxide for 3 % CID CLEAN, 7,44 g/m² ethanol for pure ALCO CID A and 2,82 mg/m² glutaraldehyde for 3 % Biocomplet plus (n=2).
To conclude, still a lot has to be investigated in residue analysis. A residue profile must be composed for each active substance, and each disinfectant must be analyzed on their residue quantity after application.
 
Samenvatting eindwerk 2011-2012: Onderzoek naar een gestandaardiseerde methode om de biologische afbreekbaarheid van synthetische detergenten en desinfectantia te bepalen
De synthetische detergenten en desinfectantia van CIRLAM worden momenteel extern getest op de biologische afbreekbaarheid. Het bedrijf zou dit onderzoek graag intern uitvoeren met een gestandaardiseerde methode. Hierbij komen er enkele moeilijkheden kijken, namelijk de keuze van het verdunningswater, de aanpak van interferenties zoals chloor, fosfaten, zware metalen ...
De biologische afbreekbaarheid wordt vaak nagegaan met de biochemical oxygen demand (BOD). Deze methode meet de biochemische zuurstofvraag bij aërobe afbraak tot koolstofdioxide, water en minerale zouten door micro-organismen. De BOD wordt bepaald met een manometrische respirometer Oxitop® waarbij de zuurstofopname wordt gerelateerd met een drukverandering. De BOD na 28 dagen incubatie (BOD28) wordt in ratio genomen met de chemical oxygen demand (COD). De chemische zuurstofvraag wordt gemeten met behulp van de Nanocolor®. Er vindt een oxidatiereactie plaats met een overmaat dichromaat en zwavelzuur gekatalyseerd door zilver. De ratio BOD28/COD moet minimum 60% zijn om te kunnen zeggen dat het product volledig biologisch afbreekbaar is.
Om de BOD te kunnen bepalen moet het product eerst worden verdund, dit om de effecten van de interfererende bestanddelen te neutraliseren en om waarden binnen het meetbereik van het toestel te bekomen. Het maken van de verdunningsreeks gebeurt met synthetisch dilutiewater die vrij is van contaminanten zoals koper, chloor, zink, chroom, lood en minerale zuren. Er wordt verdund met MilliQ water aangerijkt met nutriënten, buffer, mineralen en spoorelementen. Alvorens te verdunnen wordt eerst de theoretische zuurstofvraag berekend om een idee te hebben wat de maximale waarde kan zijn.
Er zijn heel wat interferenties mogelijk die de BOD beïnvloeden:
  • Chloor inhibeert de groei van bacteriën.
  • Metaalionen penetreren doorheen de celmembraan en beïnvloeden zo de groei van de micro-organismen.
  • Toxische stoffen veroorzaken lethale of sublethale effecten bij micro-organismen zodat de populatie vermindert en er minder afbraak mogelijk is.
  • Fosfaten verrijken het water met voedingsstoffen waardoor er meer groei is van onder andere algen. Hierdoor wordt het water zuurstofloos.
  • Stikstof aanwezig in ammonium wordt door bepaalde micro-organismen geoxideerd tot nitriet en nitriet tot nitraat. Dit vergt zuurstof en zorgt voor een pH-verlaging.
De BOD vergt naast het verdunnen van het staal nog enkele andere voorbehandelingen:
  • Het staal moet pH-neutraal zijn om een optimale werking van de micro-organismen te verkrijgen.
  • Er moeten steeds micro-organismen worden toegevoegd aan het staal. Er wordt zestien mL Polyseed®-oplossing toegevoegd aan 164 mL standaardoplossing glucose-glutaminezuur zodat een BOD5-waarde wordt bekomen binnen de aangegeven grenzen.
  • Er wordt telkens nitrificatie-inhibitor toegevoegd, twintig druppels allylthiourea per liter staal, om te voorkomen dat de micro-organismen het aanwezige stikstof oxideren en zo zuurstof verbruiken.
Een COD-meting vergt minder voorbehandelingen. Er is enkel chloor die als interferentie wordt aanschouwd. Het chloorgehalte mag maximaal 1500 mg/L zijn. Indien er een te hoge chloorconcentratie aanwezig is, wordt het staal verdund met MilliQ water. Ook de theoretische waarde wordt berekend om te verdunnen rekening houdende met het meetbereik van de COD-buisjes. De COD wordt uitgevoerd met de hoogst mogelijke concentraties om een nauwkeuriger beeld te verkrijgen van hoeveel er wordt afgebroken.
