Samenvatting eindwerk 2011-2012: Study of in vitro anticancer activity of the jujube seed extracts against the HCT 116 colorectal cancer cell line
Early studies have proven that some compounds from the jujube fruit, have an anticancer activity. The aim of this study was to investigate the anticancer activity of the compounds from the seeds of the jujube. For this experiment two types of jujube were used, an old/ripe jujube and a new/unripe jujube. To get the compounds out of the seeds, the extraction technique maceration was used. During the maceration 6 different solvents were used. In this study the extraction started from the fifth solvent. The initial four maceration steps with respectively water, ethanol, dichloromethane and ethyl acetate were already done by the lab. Only the chloroform and hexane maceration were performed. After extraction, anticancer activity was tested with a cytotoxicity test and a neutral red reuptake assay. The type of cell death was determined by DAPI staining and imaging with a fluorescent microscope. 
After maceration, four extracts were obtained: extracts from old and young jujube with chloroform solvent and extracts from old and young jujube with hexane solvent. The extracts were added in different concentrations to a HCT 116 colorectal cancer cell line and the IC50 was calculated. The two extracts from the hexane maceration gave almost no cell death, so the IC50 was not calculated. The extract from the seeds of the young jujube from the chloroform maceration gave an IC50 of 779 µg/ml. The extract form the seeds of the old jujube from the chloroform maceration gave an IC50 of 290 µg/ml. Cells can die due to apoptosis or necrosis. Cells that die of necrosis, can cause an inflammatory reaction to healthy neighbouring cells, that is why apoptosis is the preferred type of cell death during this research. The DAPI staining proved that the chloroform extract from the seeds of the old jujube makes the cells die by apoptosis. Of the four extracts, only the one from the old jujube with chloroform maceration gave a good IC50 and let the cells die of apoptosis. However there is more additional research necessary to obtain more information about the specific compounds in the extract.
